Clinical Study of Diphenpyramide on Arthritis



Abstract
Diphenpyramide(alpha-pyrydyl-biphenydyl-acetamide) is a non-steroidal non-acidic anti-inflammatory drug with analgesic, antipyretic, and uricosuric properties recently synthesized in Zambeletti Research Laboratories, Italy.
It has been carried out an open clinical study of Diphenpyramide for the drug effectiveness and its tolerance on 50 patients at Department of Orthopedic Surgery, Seoul National University Hospital between July 1983 and December 1983.
Among 50 patients, 35 patients who were followed for more than 4 weeks were included in this study; 22 patients were suffering from rheumatoid arthritis, 6 were degenerative arthritis, 3 were osteitis condensans ilii, and 3 were other orthopedic diseases. The follow up period was ranged from 4 weeks to 5 months with a mean of 5.5 weeks and following results were obtained.
1. The effect of Diphenpyramide on arthritic patients was excellent in 14 patients (40.0%), good in 15 patients (42.8%), fair in 13 patients (8.6%), poor in 2 patients (5.7%), and none in 1 patient (2.9%). 29 patients (80.8%) were better than good result.
2. There was no harmful effect on hematopoietic, renal, and hepatic function after administration of Diphenpyramide in this study. There were significant decrease of ESR and CRP, slight decrease of ASO titer, and there was no change in RA factor after treatment with Diphenpyramide.
3. Indigestion with gastric irritation was observed in 3 patients (8.6%), but the symptom was relieved with discontinuation of medication.
4. The result of this study showed that Diphenpyramide had a good anti-inflammatory effect and was well tolerated with 3 tablets(750mg) per day in adults.
